Effects of Indol-3-Carbinol on Tight Junction Activity in Human Prostate Carcinoma Cells

|
Indol-3-carbinol (I3C) is a compound found in cruciferous vegetables such as cabbage, cauliflower, broccoli and brussels sprouts. In the present study, we investigated whether I3C inhibits the cell motility, progression and metastasis of human prostate carcinoma cells. Our results indicated that I3C inhibited the viability of LnCap and PC3 cells in a dose- and time-dependent manner. Using the wound healing experiment and cell invasion study, I3C inhibited the cell motility and invasiveness of DU145 cells, but the transepithelial electrical resistance (TEER) values were increased in cells grown in transwells treated with I3C. Claudin-3 and claudin-4 have been shown to be overexpressed in prostate cancer cells and, in particular, in metastatic prostate cancer cells. But I3C occurred concurrently with a significant decrease in the levels of the proteins, claudin-3 and claudin-4 in prostate cancer cells. This result suggests that I3C may be expected to inhibit cancer progression and metastasis by restoring tight junction and their composite protein in prostate cancer.
